Cloning and Expression Analysis of Medium Chain Fatty Acids Receptor GPR84 Gene in Dairy Goat (Capra hircus)

The aim of this study is to clone medium chain fatty acids receptor GPR84 gene and analyze the ex- pression of GPR84 in different tissue and different lactation period, which lay a foundation for further understand- ing the physiological functions of dairy goat (Capra hircus) GPR84. Based on the published nucleotide sequence of bovine, human and mouse GPR84 gene in GenBank, a pair of specificity primers was designed. Coding sequence (CDS) of GPR84 was amplified by RT-PCR. Real-time quantitative PCR was used to detect the tissue specificity of dairy goat GPR84 gene. The results showed the coding region length of GPR84 gene was 1 191 bp, coding 396 amino acids. The homology analysis of dairy goat GPR84 gene indicated that the homology of GPR84 of dairy goat was found to be 96%, 88% and 82% compared with that of bovine, human and mouse. The expression analysis of GPR84 gene revealed that the mRNA expression of goat GPR84 was highest in spleen, followed by the intestine, stomach, adipose tissue, lung and mammary gland. The minimal expression was observed in muscle, liver, kidney and heart. The mRNA expression levels of GPR84 in mammary gland from dairy goat in different lactation periods analysis showed that GPR84 was expressed at a higher level at the peak lactation stage and mid-lactation stage than other detected stages (P<0.05). The lower mRNA expression of GPR84 was found at the early-lactation and late-lactation stages. In conclusion, our study indicated that GPR84 may have an essential physiological effect on mammary gland of dairy goat during lactation stages.
